Act 3 of Slay the Spire 2 is always Glory, and its boss is one of Queen, Test Subject and Aeonglass. Doormaker was removed from the game in the 18 June 2026 update and replaced by Aeonglass.

Test Subject revives twice, so the fight takes 600 damage in total (636 from Ascension 8). Doom only removes it after its second revive.
Which boss you meet
In a standard run, until you have fought all three, act 3 uses the first one you have not seen, in this order:
- Queen
- Test Subject
- Aeonglass
After that, the boss is random each run.
Ascension 10: two bosses
At Ascension 10 act 3 ends with a second boss, picked at random from the two act 3 bosses you are not already fighting.
In co-op
Boss HP is multiplied by the number of players and by 1.3 in act 3 boss fights: ×2.6 for two players, ×3.9 for three and ×5.2 for four. Queen at 2 players has 1040 HP. See co-op scaling.
